Today's unmet needs

Topical treatment of UC and IBD

Rectal gels and enemas based on hyaluronate or polysaccharides wash out quickly with defecation, pushing patients toward systemic steroids or TNF inhibitors with well-known systemic toxicity.

Oral mucositis (OM)

OM occurs in 40–80% of patients on chemotherapy or head-and-neck radiation and disrupts eating, causes severe pain, and often interrupts therapy. Topical lidocaine rinses and barrier gels persist only for 1–2 hours.

Post-ESD / EMR ulcers

Endoscopic mucosal dissection leaves ulcers prone to bleeding and perforation. Today's practice leans on systemic PPIs because reliable local depot delivery is missing.

Post-op local therapy for gastric / esophageal cancer

Combination local chemotherapy and immunotherapy for advanced GI cancers are of interest, but no carrier reliably retains drugs at the lesion under endoscopic delivery for long enough.

can do

Designed for 24+ hour mucosal retention

DOPA chemistry is rooted in the SFA measurement of Mfp-3 on mica (W ≈ 3×10⁻⁴ J/m², Lin et al. 2007) and later adhesion studies of polydopamine and PVA-DOPA films have reported mucoadhesion in the tens-of-kPa range. The Novelglue DDS platform targets 24+ hour retention against saliva, bolus and peristalsis. Novelglue-specific numbers are reported in Kawakami 2025.

EGFR-targeted delivery

A design with an EGF-fused FP2 cassette supports selective binding to EGFR-expressing epithelium and tumor tissue, exploiting the same EGF-dependent cellular responses characterized in intestinal biology (Fujii 2018 and later work).

Endoscope-compatible formats

Formulatable as gel, film or viscous liquid that delivers through endoscope channels and stays in place. Applicable to post-ESD ulcers, rectal UC, oral mucositis and more.

Excipient × drug hybrid design

Small molecules, biologics, or mRNA can be loaded or covalently fused to the FP121 core to convert existing drugs into local DDS. Designed with the FDA Novel Excipient Review Pilot Program in mind.

Fully-topical UC regimen

Reliably retaining 5-ASA, steroids or biologics at inflamed rectal mucosa could elevate remission rates while avoiding systemic steroid / biologic exposure.

OM prevention and healing acceleration

A 24-hour protective coating applied pre-treatment could reduce OM severity and chemotherapy interruptions, improving QoL during cancer therapy.

Post-ESD ulcer healing platform

Local PPI / growth factor / hemostatic deposition on the resection bed could reduce rebleeding and perforation, shortening admissions and shifting ESD toward ambulatory care.

Local immunotherapy carriers

Retention of immune checkpoint inhibitors, cytokines or mRNA vaccines at the tumor bed could reach strong local immunity at lower doses than systemic delivery.
